LAB 4 -TLC OVERVIEW: Colored Samples: 1. Get small amounts of the four standards. 2. Determine the amount of sample to be spotted by applying different number of spottings of the same standard on the same plate and developing it in 3:2 mixture of hexane:acetone (supplied). 3. Spot your unknown and the colored standard compounds on a plate, and develop it in 3:2 hexane:acetone solvent. 4. Do a confirmatory plate using selected standards and the unknown, and calculate the Rf values for all the spots. Analgesic Drugs: 5. Dissolve your unknown analgesic in 1 mL of 1:1 methylene chloride:methanol mixture. 6. Get small amounts of four analgesic standard compounds. 7. Spot your unknown solution and the analgesic standard compounds on a plate, and develop it in 15:1 methylene chloride:acetic acid mixture. Observe the plate under UV light and then develop it in an iodine chamber. 8. Do a confirmatory plate using selected standards and the unknown, and calculate the Rf values for all the spots.
